Staphylococcus epidermidis (S. epidermis) er ofte påvist i biofilm som foråsaker infeksjoner forbundet med medisinske inplantater. Noen furanoner minsker biofilm dannelse tilsynelatende uten å forårsake irritasjon eller genotoksiske effekter, og uten å påvirke S. epidermidis vekst. Furanoner hindrer kommunikasjon mellom bakterier og har stor potensial for å hemme biofilm dannelse.
Mål: Å studere biofilmhemmende og genotoksisk effekt av ulike furanoner
Materialer og metoder: 11 furanoner ble undersøkt vha bioluminisens og biofilm assayer. MIC (minste inhiberende konsentrasjon) for disse furanonene ble bestemt for å undersøke om dannelse av biofilm eller bakterievekst ble påvirket. Effekter av furanoner på bakterie kommunikasjon ble også undersøkt ved bioluminisensmålinger. Preliminære genotoksiske effekter hos CD-1 muselever og nyrer ble undersøkt vha mikromatriser og membran matriser.
Resultater: Fire furanoner ble valgt ut for videre biofilm analyser etter bioluminisens test. Hver furanon ble testet med konsentrasjoner som ikke påvirket bakterie proliferasjon, men biofilm dannelse ble signifikant redusert ved bruk av disse furanoner. For de to mest effektive furanoner (F202 og F202) som også ble testet in vivo, ble det ikke påvist akutt toksisitet eller genotoksisitet.

The coagulase-negative staphylococci (CNS) are commensals of human skin and mucous membranes. Staphylococcus epidermidis is the most frequently isolated CNS species, and also one of the major nosocomial pathogens associated with infections on medical implants. The cause of infections is primarily related to biofilm formation on the implant. S. epidermidis infections represent a major burden to the public health system, and are often responsible for prolonged hospitalisation and occasional deaths. Microorganisms in nature prefer a biofilm life style. Biofilm infections are often difficult to treat due to increased resistance both to antibiotics and to the immune system. The algae Delisea pulchra produces bromated furanones that prevent bacterial biofilm formation on its surface. Similar furanones may interfere with both autoinducer-2 and autoinducer-1 signalling in Gram-positive and Gram-negative microorganisms without
affecting bacterial growth.

Bacteria communicate using small signaling molecules as part of a system of communication called quorum sensing (QS) to control gene expression for synchronized bacterial behaviors. A task performed by bacteria that is often controlled by this means is biofilm production, making bacteria more resistant to external factors. Naturally occurring furanones that were isolated from a red macroalgae have been shown to have the ability to interrupt this communication. As a result thiophenones have been synthesized and investigated as a novel class of quorum sensing inhibitors (QSIs). Molecules of this class have greater biofilm reduction abilities than furanone equivalents for some bacteria. This discovery resulted in many more thiophenones with a variety of functional groups being synthesized using both classical and new methods. These compounds have been used in biological assays to determine their quorum sensing inhibition (QSI) potential, and have shown promising results. In this study tiobovolide has been synthesized and confirmed to exhibit some QSI properties. In addition to this trifluoromethyllated thiophenones have been synthesized and tested for QSI ability. It was found that most exhibited QSI properties, but that compounds with methyl groups in the 3- and 4-position showed no biological activity. This finding may support a 1,6-Michael-type reaction mechanism that has been suggested to be responsible for bioactivity.
